Description
Vita-D (300,000 IU) provides cholecalciferol at a 300,000 IU dose per 1.5 mL ampoule, representing a higher single-dose depot injection for rapid correction of severe vitamin D deficiency. The oil-based or aqueous cholecalciferol depot releases gradually from the intramuscular injection site, sustaining elevation of serum 25(OH)D levels over weeks to months following a single administration. Indicated for patients with severe deficiency (serum 25[OH]D < 20 ng/mL) or malabsorption syndromes where oral supplementation is unreliable.

Protocol
Administer 300,000 IU (1.5 mL) by deep intramuscular injection into the gluteus maximus using a 23G 38 mm needle with aspiration; a single loading dose is typical for severe deficiency. Reassess serum 25(OH)D at 12 weeks; if levels remain below 30 ng/mL, consider a second dose. Monitor serum calcium and renal function; use caution in patients with granulomatous conditions or primary hyperparathyroidism.
